Por favor alguien me podría explicar como usar esta question phrase...
¿Como me doy cuenta si tengo que usar What makes osea con s y cuando sin s osea What make?
Podrían darme algún ejemplo por favor.
He leido esto: What makes you feel happy? como se que no debo poner make sin s

what makes... singular
What makes you happy?
What things make you happy?

I use it without auxiliaries because it is a Question without auxiliaries? Am I right? o What is the reason of it?

porque no uso auxiliar en la pregunta: osea porque no digo What does make you happy? EL Auxiliar DOES no lo uso o el Do... porque va directa sin auxiliar esa pregunta? osea no digo What makes do you happy?
Sino que digo directamente What makes you happy? porque no lleva auxiliar do or Does

Sí podemos usar el auxiliar para hacer hincapié.

We use the auxiliary when there is another subject besides "what."
Ex.
What did he say? / What does he say?
Here, "he" is the subject of the verb "say."
Contrast that with the following:
What gives you the right?
Here, "what" is the subject of the verb "give."
Maybe someone with better knowledge of grammar terminology can tell you what these forms are called.
